Human Norovirus (HuNoV)
Genotyping and Titration

HuNoV are classified in Genegroups (I-VI) and distinct genotypes (i.e. GII.4). For this project we will establish a RealTime PCR (qRT-PCR) to titrate HuNoV from patient samples. Genotyping will be performed by PCR amplifying a specific region in the ORF-1/2 junction and seqeuncing.

Qualification:

  • understanding viral evolution of plus sense RNA viruses
  • development and optimization of viral diagnostic techniques
  • working under BSL-2 conditions with human pathogenic agents

Determinants for Norovirus Cell Entry

Virus entry is a multi-step process involving vioral proteins (ligands) and host surface proteins (receptors). For this project, we will characterize viral and host factors that determine norovirus entry using the murine norovirus as a model organism.

Practical skills:

  • cell culture
  • advanced molecular biology techniques
  • advanced virology techniques, virus titration and purification
  • protein expression in insect cells (baculovirus expression)
  • protein purification by chromatography

Qualification:

  • understanding entry of non-enveloped viruses
  • understanding of receptor-ligand interactions
  • working under BSL-2 conditions with animal pathogenic agents
